According to PubMed, this 2003 study (Blanes-Mira et al., Biochemical Journal, DOI 10.1042/BJ20030509) screened an alpha-helix-constrained combinatorial peptide library against detergent-permeabilized bovine chromaffin cells and rat hippocampal neuron cultures, identifying peptides that block SNARE-core-complex assembly and inhibit Ca2+-evoked catecholamine and glutamate release. This established the in vitro exocytosis-inhibition pharmacology model used to characterize SNARE-mimetic peptides in the same structural class as SNAP-8.

According to PubMed, this 2002 study (Blanes-Mira et al., International Journal of Cosmetic Science, DOI 10.1046/j.1467-2494.2002.00153.x) characterized Ac-Glu-Glu-Met-Gln-Arg-Arg-NH2 (Argireline), the parent hexapeptide sequence from which SNAP-8 was later derived by C-terminal extension. In vitro exocytosis assays showed the hexapeptide interfered with SNARE-complex formation and stability and inhibited neurotransmitter release with potency comparable to botulinum neurotoxin type A, establishing the SNARE-inhibition mechanism this peptide class is researched for.
